I know many have put the XB heads on Tube motors, but does anyone have experience with XB cylinders (or new Sportster cylinders) on a Tuber? I like the idea of greater finning and spine locks (even if I have to make alterations in order to have room to fit the Mikuni). I plan to replace the cylinders anyway with newer spine lock, but why not also get some added finning.

OK
You can use the stock Cyl with the XB heads OR go with the XB cyl if ya like the whole top end is interchangeable.

And you will not have a problem with teh new mikuni fitting on there.

You will however need to drill mounting holes in the heads for mounting the carb bracket as the XB heads are blank there.

Or get XL1200R heads.
Same as XB heads, but have the carb mounting.

It is possible you may have some interference with fins on the XB cylinders and the Mikuni.I had to do a bit of grinding on My S-2 as carb body hit a bit.And the gas line between the cylinders is a squeeze now.XL1200R cylinders are the way to go.
